Hyperbaric oxygen cut body weight 9–22% — in high-fat-fed mice, alongside an immune injection

Mice given hyperbaric oxygen (HBO) therapy and an IL-2/anti-IL-2 complex immune treatment lost 9–22% of their diet-induced body weight, an effect that was independent of caloric intake. Area under the curve fell by 20–25% on glucose tolerance testing and 30–35% on insulin tolerance testing. The subjects were male C57BL/6 mice, not humans, and no significant changes appeared in the low-fat-diet groups — nothing improved in animals whose metabolism was already normal.

Hyperbaric chambers are sold as recovery equipment. This study asked what the exposure does to body weight and glucose handling in animals. Male C57BL/6 mice were fed a low-fat or high-fat diet for 16 weeks while receiving an IL-2/anti-IL-2 complex (IL-2C), hyperbaric oxygen, or both. Each treatment is known to expand regulatory T cells; their combined effect on obesity had not been characterised.

What the numbers moved

  • Body weight — a 9–22% reduction in diet-induced weight, independent of caloric intake.
  • Glucose — intraperitoneal glucose tolerance test AUC down 20–25%.
  • Insulin sensitivity — intraperitoneal insulin tolerance test AUC down 30–35%.
  • Adipose tissue — reduced adipocyte hypertrophy and fewer crown-like structures (immune cells surrounding dying fat cells) in epididymal fat.
  • Immune profile — CD4+FoxP3+ regulatory T cells restored, pro-inflammatory M1 macrophages and HIF-1α expression suppressed.
  • Low-fat-diet groups — no significant change.

In brown fat (iBAT), multilocular adipocytes — cells holding many small lipid droplets — were restored, and in subcutaneous white fat (iWAT) adipocyte hypertrophy was reduced. The combination produced the most consistent improvements.

Can this be carried over to humans?

Three things get in the way. First, the species. Mouse metabolic responses are not human ones, and there is no human data here. Second, this was not a standalone therapy: the most consistent results came from HBO combined with an immune treatment, and IL-2C is a categorically different intervention from sitting in a chamber. Third, and most importantly, nothing happened in the low-fat-diet mice. What the treatment changed was a state already damaged by a high-fat diet; it did not make a normal state better.

The practical read for a lifter

The pattern here repeats across recovery equipment. When a mechanistic finding is carried into marketing copy, the species, the co-treatment and the baseline condition disappear with it — the same structure as the study where melatonin prevented muscle atrophy and the subject was a cell culture. Checking the order of operations beats buying hardware: sleep came before the altitude chamber, and a recovery routine does not improve by addition.

Hyperbaric oxygen therapy is a medical treatment with defined indications and reported adverse effects. This was an animal study and cannot serve as human evidence for using hyperbaric oxygen for weight loss or glucose control.
